Perbedaan Botol Kultur terhadap Pertumbuhan Eksplan Anyelir Hiperhidrisitas

Hyperhydricity is an important problem in carnation tissue culture. Fail in overcoming the problem can influence the succesful of plantlet acclimatization. Hyperhydritized-plantlets are normally difficult to be acclimatized and easy to become wilting and die. The aim of this study was to know the effect of different types and sizes of culture vessel in reducing hyperhydricity. The experiment was conducted at Tisue Culture Laboratory, IPPT, Faculty of Agriculture, Universiti Putra Malaysia from Juni to November 2002. Six types different in sizes used in the study are tube vessel, erlenmeyer 100,150,250,500ml, and GA7 vessel. Completely randomized design with four replications was applied in this research. Result of this study showed that tube vessel was to most appropriate culture vessel to grow hyperhydritized-eksplant reverting to normal growth of them was indicated by the lowest percentage of hyperhydricity (42%), the highest leaf clorophyll content (0,1377mg/mg), the lowest water content (89%), and the highest dry weight of biomass (11%) with good quality of shoots compared to others.
